Brightfield
Histopathology
using Spectral Absorption and
Histogram Functions
"Characterizing
any sample in absorption or
% reflection provides absolute
measurements that conveys a host of information. When
combined with a histogram we have powerful statistics"

Plots show the absorption spectra of areas in circles

Absorption
measurements on micro-areas in Figure
1 shows a stained keratinocyte that was illuminated
in brightfield. Absorption spectra, measured in optical
density, are shown in the plots alongside.

| Figure 2. Shows
the spectral histogram of all classified spectra that
correlated with library spectra. |
Spectral
Histograms
Spectral ratios present in either the entire field of view (FOV), or local
regions, can be presented in histogram format.
Because natural
products tend to present significant local heterogeneity
presenting the results of a spectral classification in
histogram format tends to even out local anomalies and
provide an accurate statistical overview.
